Barcode list format (tab- or space-separated table)
Fields cannot contain spacesRequired fields (in order):
- Barcode name - Unique identifier for the barcode
- Barcode type - Type of barcode (e.g., i5, i7)
- Barcode sequence in physical oligo - The actual barcode sequence
- Physical primer/adapter backbone sequence - Backbone sequence where N characters indicate barcode position
- Orientation of physical barcode in final library - Either "forward" or "reverse"
Example:
Barcode name Barcode type Barcode sequence in physical oligo Physical primer/adapter backbone sequence (N characters indicate barcode position) Orientation of physical barcode in final library P7_A01 i7 CGGTTCAA aaaaaaaaaaaaaaannnnnnnngtgtgtgtgtgtgtgtgtgtgt reverse P7_A02 i7 GCTGGATT aaaaaaaaaaaaaaannnnnnnngtgtgtgtgtgtgtgtgtgtgt reverse P7_A03 i7 TAACTCGG aaaaaaaaaaaaaaannnnnnnngtgtgtgtgtgtgtgtgtgtgt reverse
Plate Coordinates: If the barcode name ends with a plate coordinate pattern (e.g.,
_A01,-B12), the coordinate will be automatically extracted and stored. The pattern is:[_-][A-H]\d{1,2}where the letter represents the row (A-H) and the number represents the column (1-12). Examples:P7_A01,P7_A02,P7_B12,P5-H8
